摘要:鹿茸是我国传统名贵中药,具有广泛的药理学活性。鹿茸作为一种特殊的软骨,是一个天然的细胞因子库,其中含有大量的蛋白质及蛋白多糖。近年来大量研究证明,蛋白多糖具有抗肿瘤作用,而以往对鹿茸的研究主要是醇提物,其中蛋白质和多糖类水溶性物质研究较少,对其水溶性物质进行系统的抗肿瘤研究更是鲜见。本试验调查了具有抑制作用的顶端鹿茸(TDVA)对前列腺特异性抗原(PSA) 相关基因在人类前列腺癌细胞LNCaP中的表达和迁移的影响。TDVA能抑制前列腺特异性抗原的表达;半径试验表明TDVA能抑制前列腺癌细胞的迁移行为。经过鹿茸作用后基质金属蛋白酶(MMP)9和血管内皮生长因子(VEGF) 的表达也减少。相反,TDVA促进了组织金属蛋白酶抑制(TIMP)1和 (TIMP)2的表达。研究结果表明,TDVA具有对前列腺癌细胞LNCaP的抗转移活性。本研究为鹿茸抗肿瘤作用的机制研究奠定了基础。

Abstract: Deer velvet antler is a kind of rarely traditional medicine, and it has extensive effect on curing disease. As a special cartilage, velvet antler is a natural cytokine ‘storeroom’, and has lots of protein and proteoglycan. It is indicated that proteoglycan has good effect for antitumor lately. Previous research on velvet antler is more involved in ethanol extraction mostly than in protein and polysaccharide which are dissolved in water, and their roles in antitumor have not been investigated systematically. In this study, we investigated the inhibitory effect of top DVA (TDVA) on the expression of prostatespecific antigen (PSA) and migrationrelated genes in the human prostate cancer cell, LNCaP. The TDVA downregulated the expression of PSA. In addition, the RadiusTM assay revealed that TDVA inhibited the migration behavior of prostate cancer cells. Furthermore, the expression of matrix metalloproteinase (MMP)9 and vascular endothelial growth factor (VEGF) was also decreased with TDVA. On the contrary, TDVA increased the tissue inhibition of metallo proteinase (TIMP)1 and (TIMP)2. Taken together, our findings indicate that the TDVA possesses antimigration activity on prostate cancer cells. This study provides a basis for investigation of antitumor.mechanism by velvet antler.
