3本试验旨在探讨功能性氨基酸N对肉鸡肝脏脂代谢的影响及机理,实验选用72只重量均等的1日龄立华优麻肉仔鸡,随机分为6组,每组12只。分为基础日粮组(对照组,CON),基础日粮加0.1%功能性氨基酸N添加组(N2组),基础日粮加0.2%功能性氨基酸N添加组(N4组),按标准程序饲喂至28日龄时,以上各组鸡平均分成2个组,其中一组皮下注射皮质酮,另一组注射等量的溶剂作为对照。从而将实验动物分为以下6组基础日粮组+溶剂组(对照组,CON),基础日粮+皮质酮注射组(T组),添加0.1%功能性氨基酸N+溶剂组(N2),添加0.1%功能性氨基酸N+皮质酮注射组(N2T组),添加0.2%功能性氨基酸N+溶剂组(N4),添加0.2%功能性氨基酸N+皮质酮注射组(N4T组),连续7天注射。35日龄时屠宰采样。结果表明,与CON组相比,T组脂肪肝症状极为显著,脂滴增大、血液和肝脏中TG含量升高(P < 0.01);日粮中添加功能性氨基酸N能显著缓解肉鸡脂肪肝现象,且高剂量组的作用更加明显。与单纯的皮质酮注射组相比,N4T组肝脏中固醇调节元件结合蛋白-1(SREBP1)、硬脂酰辅酶A去饱和酶(SCD)、脂肪酸合成酶(FAS)

及乙酰辅酶A羧化酶(ACC)的基因表达量显著降低(P< 0.05),而脂肪酸β氧化相关酶如肉碱脂酰转移酶I(CPTI)、过氧化物酶体增殖物激活受体α(PPARα)的基因表达量显著增加(P< 0.05)。因此,本实验结果提示:皮质酮连续注射诱导肉鸡脂肪肝的发生,而功能性氨基酸N通过下调肝脏脂肪酸合成酶而上调脂肪酸分解氧化酶的基因表达,以缓解肉鸡脂肪肝症状。
Effect of Functional Amino Acid N on Lipid Metabolism in Broiler Chicken and Its Mechanism

Abstract:The aim of this study was to investigate the effect of functional amino acid N on liver lipid metabolism in broiler chickens. A total of 72 broilers were randomly divided into 6 groups (n = 12). The control group (group CON) and corticosterone group (group T) were fed with basal diet, low concentration 0.1% functional amino acid N feed group (group N2) and low concentration 0.1% functional amino acid N feed group plus corticosterone treatment (group N2T), a high concentration of 0.2% functional amino acid N supplement (group N4) and a high concentration of 0.2% plus corticosterone treatment (group N4T). Corticosterone injection was started at the age of 28 days old, the broilers in the T group, the N2T group and the N4T group were treated with 4 mg / kg / d dose injection of corticosterone, the rest of the group received ethanol injection for 7 days. At 35 days old all broilers were slaughtered for samples. The results showed that compared with the CON group, the fatty liver of T group was significantly increased (P <0.01); and gene expression of hepatic sterol regulatory element binding protein (SREBP1), desaturase A desaturase (SCD), fatty acid synthase (FAS)and stearoylCoA desaturase (ACC) were significantly increased in corticosteronetreated group (P <0.05), while genes involved in βxidation of fatty acids in the liver including carnitine acyltransferase I (CPTI), and peroxisome proliferatoractivated receptor (P <0.05) was significantly decreased (P <0.05). Functional amino acids N supplemented into diet can obviously alleviate the fatty liver in broilers induced by corticosterone injection through regulating genes expression in the liver. In summary, the addition of functional amino acids in diets has reduced the effect of fat deposition on broiler fat.
