摘要:本试验旨在探究肠道微生物代谢产物丙酸对猪摄食相关胃肠激素分泌及相关基因表达的影响。通过体外结肠组织灌流丙酸钠试验表明,5mM和20mM丙酸钠在30 min、60min时分别能够显著刺激PYY、GLP1的分泌。通过体内盲肠瘘管灌注丙酸钠试验表明,丙酸显著提高结肠中GLP1、PYY浓度(P<0.05),而对CCK、Ghrelin、Insulin浓度无显著影响(P>0.05);丙酸显著上调结肠中GLP1R mRNA表达(P<0.05),但对GCG、PYY表达无显著影响(P>0.05);丙酸显著下调下丘脑促进摄食基因AGRP的表达(P<0.05),但对NPY以及抑制摄食基因POMC、C *好棒文|www.hbsrm.com +Q: ¥351916072$ 
ART无显著影响(P>0.05)。综上,丙酸能够刺激结肠分泌GLP1、PYY,并上调GLP1受体基因表达,下调下丘脑促摄食基因AgRP表达。本文可为从肠道微生物营养代谢角度制定猪营养调控策略提供理论指导。
The effects of propionate on the appetite and its related genes’ expression of pigs

Abstract:The purpose of this study was to investigate the effects of propionnate, which is an intestinal microbial metabolite, on the secretion of gastrointestinal hormones and related gene’s expression in pigs. With infusing sodium propionate in vitro colon tissue showed that 5mM and 20mM propionate could significantly stimulate the secretion of PYY and GLP1 at 30 min and 60min. This test showed that propionate can significantly increased the concentrations of GLP1 and PYY in the colon (P<0.05), but had no significant effect on the concentration of CCK, Ghrelin, and the Insulin(P>0.05);the expression of GLP1R mRNA was significantly upregulated in colon of propionate(P<0.05), but it had no significant effect on genes’ of GCG, PYY expression(P>0.05); propionate significantly reduced hypothalamic feeding promotes the expression of AGRP gene (P<0.05), but no significant effect on NPY and POMC, CART gene inhibits food intake (P>0.05).In conclusion, propionate can stimulate the secretion of GLP1 and PYY in colon, and up regulate the expression of GLP1 receptor gene and down regulate the expression of hypothalamic feeding gene AgRP. This article can provide guidance in pig breeding from the point of nutrition metabolism.
